For some reason twice now in a week I open vdj everything is set music plays everything is on the screens of the 4500 but non of the buttons will respond. Any ideas?

Do you have the latest firmware installed from http://www.denondj.com ?

I've had that happen a couple of times.
Is your 4500 mounted in a case or free standing? If so make sure the cables and where they connect to the 4500 are free of obstructions and protrusions that may jar or loose the cable. If the usb cord gets bumped or loosened even a little it could give you the same result. Had that happen to me during a set.
Do you plug your 4500 into the same usb port every time? My old laptop got picky towards the end as to where I could plug things.
Also check the Hardware Technical Support Page. There's a spot for Denon products...alot of useful info there.

I've been using the HC4500 for a few years now and what I have notice is that if I leave VDJ running but not playing (about an hour or more) when I come back the controller does not respond but VDJ still works. I thought at first it was something with USB power management or something but I've change the settings. This doesn’t really bother me but it's worth mentioning in this topic.

I have not checked for the latest firmware, but I am using VDJ 7.0.2. I always plug it into the same USB port and it is mounted in a case and stays in there for good. I will check for the latest firmware and try that.
