Could try killing all the bass and the treble - vocals are normally mid-range frequencies...

If want to try and make an acapella, then you ideally need to find an instrumental version of the same tune, load both into an audio editing program (i.e. Audacity) then invert the phase of the instrumental and run them together.

The easiest method is to purchase the proper acapella of the song, if available.
